He and his wife lived on Broad River at Fish dam, and he is buried in Fishdam Cemetery, SC. He served in the 6th VA Regiment in the Revolutionary War with George Washington at Yorktown. He was a wealthy businessman, owned many acres of land in the Fishdam area, and he and his business partner, James Thomas, operated a ferry at Fish Dam Ford for many years. In 1791 the Union County Court "ordered that Spilsby Glenn be the overseer of the road from the Fish Dam ford on Broad River to the Ninety Six Road that leads to Hamilton's Ford on the Tyger River to Cain Creek and that he keep the same in good repair." It was Spilsby and his father Nathan who gave the land for the Fish Dam Meeting House, one of the first Methodist churches in the area. In 1843, he granted two acres around the Fish Dam Meeting House for a burial ground. By the time that Spilsby and Sarah Glenn married, all of the Glenns were attending the Methodist Church, since there were no Episcopal Churches in the South Carolina "back country," and they were all staunch supporters of Methodism. He was appointed as one of the Trustees in the parsonage of "Glenn's Chapel" in 1818.
